RNA酶保護試驗((RNase Protection Assay,RPA)是通過液相雜交的方式,用反義RNA探針與樣品雜交,以檢測RNA表達的技術。1。原理:雙鏈RNA(雜交的)能夠抵抗RNA酶的降解。2。應用:檢測RNA表達3。與Northern雜交和RT-PCR比較,RPA有以下幾個優點:1. 檢測靈敏度比Northern雜交高。由于Northern雜交步驟中轉膜和洗膜都將造成樣品和探針的損失,使靈敏度下降,而RPA將所有雜交體系進行電泳,故損失小,提高了靈敏度。2. 由于PCR擴增過程中效率不均一和反應“平臺”問題,基于PCR產物量進行分析所得數據的可靠性將下降,而RPA沒有擴增過程,因此,分析的數據真實性較高。3. 由于與反義RNA探針雜交的樣品RNA僅為該RNA分子的部分片段,因此,部分降解的RNA樣品仍可進行分析。4. 步驟較少,耗時短。與Northern雜交相比,省去了轉膜和洗膜的過程。5.
