ELISA實驗結果為什么有時候會復測？
在ELISA實驗過程中時常會出現復測現象即我們時常說的"花板"現象，這種現象影響檢驗結果的正確判讀，對工作造成一定的不利影響，一旦出現“花板”，實驗結果必須放棄，重新進行，不僅浪費物料和時間，而且還會出現樣本量缺少、交叉污染、報告延遲等一系列問題。影響因素分析如下：一、標本因素正確處理標本，防止大規模溶血、血漿層異物、使用一次性加樣槍頭，防止交叉污染。血清和血漿均可以用于檢測，但血清的分離應在抽血后等血液完全凝固后再離心。二、試劑因素正確保存及使用有效試劑。使用過期試劑及底物接觸金屬容器、底物混合后放置時間較長均影響實驗結果的準確性。選擇試劑時應選擇靈敏度高、特異性好、穩定性好，批間差小的試劑。不同廠家的試劑不能混用，包括底物和洗滌液。檢測前應將試劑放置室溫平衡半小時，洗滌液應現用現配，同時觀察濃縮洗滌液中有無結晶，若有結晶應放37℃水浴中融化后才可使用。三、技術人員操作因素 嚴格按照相關實驗SOP......閱讀全文
